Tyrosinase plays a key role in melanogenesis, as it controls the rate of melanin deposition. 4-Butylresorcinol has an inhibitory effect on melanogenesis, by directly inhibiting tyrosinase activity, as well as inhibiting tyrosinase synthesis in B16 melanoma cells, without causing any cytotoxicity. In several in vitro studies, 4-butylresorcinol was shown to inhibit melanogenesis, as well as tyrosinase activity and TRP-1.
Mechanism of action
1 Interferes with tyrosinase synthesis and glycosylation prior to melanin synthesis, preventing enzyme uptake by melanosomes
2 During melanin synthesis, inhibits enzyme activity, acts as a competitive inhibitor of tyrosinase and TRP1 enzymes, and reduces the formation of by-products that promote melanogenesis.
3 After melanin synthesis, enhances tyrosinase degradation, inhibits the transfer of melanosomes to keratinocytes, and has a photoexfoliation effect due to fatty acids, a function enhanced by the presence of fatty acids, and regulates the degradation of tyrosinase.
